چکیده

Measures of immune function in birds, while routine in traditional areas of pathology and physiology of domestic fowl (e.g. Goto et al. 1978; van der Zijpp 1983; Laudert, Sivanandan & Halvorson 1993;Chu et al. 1995), have now been recognized as valuable tools to study behaviour, ecology and toxicology of wild species (e.g. Lochmiller, Vestey & Boran 1993; Dufva & Allander 1995; Saino, Calza & Møller 1997; Svensson & Skarstein 1997; Moreno et al. 1998). Variables related to immune function such as differential and total white blood cell counts, and production of antibody-producing lymphocytes against sheep red blood cells (SRBC) are commonly used but require specialized training or laboratory facilities. An inexpensive, easily applied skin test using phytohaemagglutinin (PHA) as a mitogen, has become a popular, effective test that can be applied with minimal training and without specialized equipment. The skin test provides a measure of the proliferative response potential of circulating T lymphocytes to an injected mitogen. Phytohaemagglutinin has long been recognized for its mitogenic and blastogenic properties (Hungerford et al. 1959). This test has been routinely applied, with some variation in the body sites used, according to a long-established protocol developed in poultry science (Goto et al. 1978). In the seminal work of Goto et al. (1978), histological assessment of skin that had been injected with 75 μg PHA revealed an infiltration by heterophils within 3 h. The transient heterophilia, a non-specific inflammatory reaction to mild antigenic stimulus, was no longer evident 6 h after injection. The main cellular response, observed at 6–12 h after injection, consisted of a prominent perivascular accumulation of Functional Ecology 1999 13, 567–572

The PHA Test as an Indicator of Phagocytic Activity in a Passerine Bird

Several techniques in ecological immunology have been used to assess bird immunocompetence thus providing useful information to understand the contribution of the immunological system in life-history decisions. The phytohaemagglutinin (PHA)-skin test has been the most widely employed technique being interpreted as the sole result of T lymphocytes proliferation and hence used to evaluate acquire...

Nestling immune response to phytohaemagglutinin is not heritable in collared flycatchers.

The response to intradermally injected phytohaemagglutinin (PHA-response) is a commonly used quantification of avian immunocompetence (the ability to resist pathogens). Parasite-mediated sexual selection requires heritable immunocompetence, but evidence for heritability of PHA-response in birds largely stems from full-sib comparisons. Using an animal model approach, we quantified the narrow-sen...

Effects of repeated comparative intradermal tuberculin testing on test results: a longitudinal study in TB-free red deer

BACKGROUND Diagnosing tuberculosis (TB) in farmed red deer (Cervus elaphus) is challenging and might require combining cellular and humoral diagnostic tests. Repeated skin-testing with mycobacterial purified protein derivatives (PPDs) might sensitize or desensitize the subjects to both kinds of diagnostic tools. We evaluated the effect of repeated (every 6 months) comparative tuberculin skin te...

Effects of T-cell-mediated immune response on avian oxidative stress.

We evaluated the oxidative cost paid by birds when coping with an immune challenge. We used the phytohaemagglutinin skin test (PHA) to assess the effects of the T-cell-mediated immune response on the concentration of reactive oxygen metabolites (ROMs), total antioxidant barrier (OXY) and total serum carotenoid concentration in wild nestlings of the Eurasian kestrel (Falco tinnunculus). Immunost...

Tuberculin, Tetanus Immunoglobulin and DPT Vaccine as an Avian in vivo T- Lymphocyte Mitogens

The avian phytohaemagglutinin skin test is being proved as an in vivo system for the evaluation an avian in vivo T cell mitogenicity. The test system was one week old Gallus domesticus broiler Chickens. Five replicates were done for each of the whole, 1:10 dilutions of each of 0.05 IU tuberculin, tetanus immunoglobulin and DPT vaccine as test materials. The evaluation parameters were the skin i...
